カドヘリン活性化に必要なα-カテニン機能領域の同定とその結合分子のクローニング

研究概要	カテニンは、カルシウム依存性の細胞接着分子カドヘリンの構造と機能の解析中に申請者らにより発見された分子で、カドヘリンの細胞質ドメインに結合していて、カドヘリンの機能発現に必須である。カテニンのうち、β-カテニンあるいはγ-カテニンは直接カドヘリンの細胞質ドメインに結合し、カドヘリンとα-カテニンをつなぐアダプターとして機能している。α-カテニンはそのアミノ末端の領域でβ-カテニンあるいはγ-カテニンに結合し、さらに分子の中央部分でα-アクチニンに結合するすること、あるいはそのアミノ末端とカルボキシ末端でアクチンフィラメントに結合し、それをアクチン束にする活性が報告されている。したがって、α-カテニンはカドヘリンをアクチンフィラメントにリンクすることに関与していると考えられてきた。しかしながら、カドヘリンが細胞接着分子として機能する際に何故α-カテニンが必要とされるのか、言い換えれば、α-カテニンのどのような機能がカドヘリンの活性発現に必要なのかという問いに対する答えはいまだ得られていない。本研究ではまずα-カテニン上のどの領域がカドヘリンの活性発現に必要かを明らかにした。すなわち、α-カテニンのカルボキシ末端の1/3の部分(295アミノ酸)が必要十分であることを、E-カドヘリンとα-カテニンのキメラ分子および一連の欠失を持つ変異α-カテニン分子の発現実験により明らかにした。この領域はアクチン結合部位を含むが、それよりも77アミノ酸残基大きい。現在、この領域を使い、酵母菌のtwo-hybrid systemあるいはリガンド・ブロット法、アフィニティー精製等の方法を駆使して、この部位に結合する分子の同定ならびにそのcDNAクローニングを行っている。
