糖転移酵素のゴルジ装置停留に関わる分子種の同定と解析

研究概要

ゴルジ装置に存在する糖転移酵素は2型の膜蛋白質であり、N末側を細胞質側に、C末側をゴルジ内腔に向けている。糖転移酵素をゴルジ装置のどの位置に停留させるかを決めているメカニズムを明らかにするため、以下の2つの実験を行った。(1)酵母two hybrid systemを用いた糖転移酵素と相互作用する蛋白質の検索:糖転移酵素に対する抗体を用いた検討から、癌細胞由来の細胞株では細胞極性がみだれている可能性が認められた。このため、外科手術の際に得られたヒト非癌部右半結腸粘膜を材料としてcDNAを合成し、CAL4 Activation Domain vector pPC86に組み込みcDNAライブラリーを作成した。右半結腸粘膜では、ガラクトース転移酵素I、フコース転移酵素III(ルイス酵素)、H酵素、Se酵素が発現していた。これらの転移酵素のcoding領域全長、及び、酵素活性部位、膜貫通部位と細胞質尾部を分けてDNA Binding Domain vector pDBLuに組み込み、スクリーニングを行った。現在、いくつかの陽性クローンを得て、解析を行っている。(2)生体内における変異型ルイス酵素の解析:赤血球上や消化管におけるタイプIルイス抗原の発現は、ルイス酵素により規定されている。ルイス酵素遺伝子には、野性型と3つの変異型(le1、le2、le3)の4つの対立遺伝子がある。生体内におけるこれらの変異酵素の解析を行ったところ、膜貫通部位のLeu20 to Argの変異がle3酵素のゴルジ膜への停留を妨げていることが明かになった。このため、le3酵素は糖脂質上の抗原は合成できず、赤血球の表現型は陰性となり、正常大腸や大腸癌のムチン蛋白質上の抗原量も減少することが明かになった。
