GM-CSFによる細胞増殖制御機構の研究

研究概要

GM-CSFは多能性血液幹細胞や多くの血球細胞の増殖と分化を制御し、その除去により細胞は速やかに死に到る。我々はGM-CSFのレセプターとシグナル伝達について主にβ鎖に集中して研究を展開し、変異体レセプターの使用によりシグナル経路の選択的活性化または不活性化が可能である事を示した。本研究では増殖能のみ持ち、MAPキナーゼカスケードの活性化を伴わない種々の変異体を利用してGM-CSFレセプターからDNA合成開始に至るシグナル伝達とメカニズムについて分子生物学的および生化学的手法を用いて明らかにすることを目的とした。増殖能に必要なレセプタードメインを用いてGM-CSF刺激による未知の誘導遺伝子をサブトラクション法により複数単離したが、機能的単離法でないため単離した遺伝子の機能解析に困難が伴った。そこで生化学的な遺伝子単離と、単離された遺伝子の機能解析のため細胞の各フラクションの再構成系によるin Vitroでのリガンド刺激に依存した機能解析の系を確立し、レセプターからDNA合成開始に至るシグナル伝達とメカニズムについて明らかにすることを目的とした。すでに確立されたSTAT5の活性化を指標としたIn Vitroのリガンド依存的なGM-CSFシグナル伝達系ををc-mycの活性化に応用することを目指したがこの系は確立することはできなかった。一方でリコンビナントのレセプター部分を用いた生化学的に相互作用する分子の検索はレセプターβ鎖に特異的に結合する分子のクローニングに至った。これをさきに確立したSTAT5の試験管内系で活性測定を行ったことで遺伝子クローニングに生化学的方法を用いた試験管内系を組み合わせてサイトカインの細胞増殖に至る制御機構を検討するという当初の目標は達成された。しかしながら生化学的方法の増殖そのものの解析系への応用という点は今後の問題として残された。
