ラットより単離したRuvB類似DNAヘリカーゼTlP49の解析

研究概要	基本転写因子TBPに結合する因子(TBP-interacting protein;TlP)の探索を行い、これまでに多数のTlPを同定した。本研究ではこれらTlPの中でATPase/ヘリカーゼの特徴的なモチーフであるWalkerA、Bを持ち、かつ原核生物の組み換え因子RuvBと高い相同性を持つTlP49について詳細な解析を行った。1)自己抗原としてのTlP49;ヒトの自己免疫疾患において核内の高分子複合体が自己抗原となっている例が報告されている。抗TlP49抗体を用いてHeLa細胞を染色したところ、核がドット状に染色されたことより、TlP49は核内因子であることが明らかになった。また、多発性筋炎、全身性エリトマトーデス、リユマチ、自己免疫性肝炎の患者より得た血清中に有意にな頻度で抗TlP49抗体を検出できたことより、これらの自己免疫病で自己抗原となっているを見い出した。2)TIP49遺伝子ファミリー;TlP49遺伝子は真核生物から古細菌まで高度に保存されており、古細菌では一種類、真核生物では酵母からヒトにいたるまで二種類の遺伝子が存在した。これまで解析したラットTlP49を含むグループをTlP49a、もう一方をTlP49bと命名した。ヒトTlP49bも新規のATPase/DNAヘリカーゼであり、TlP49aと-bが複合体の状態で存在していた。二つの蛋白質は協調的に細胞内で働くと考えらる。本研究ではRuvB類似因子TlP49a、TlP49bが新規のDNAヘリカーゼであることを証明し、その他生化学的、臨床的にも多くの成果を上げた。今後、酵母に存在するTlP49ホモログを解析することで遺伝学的、細胞生物学的な解析を進めていく予定である。現在、諸外国の研究グループもTlP49の重要性を認識し、本因子が転写やクロマチンリモデリング、あるいは癌化と密接に関係することが明らかにされつつある。本因子は、核内でのDNAトランスアクションや核ダイナミックスを解明する手がかりを与えるものとして期待される。
