ゼブラフィッシュ突然変異体を用いた中枢神経の背腹軸形成機構の解析

研究概要	頭部腹側でも発現するoep遺伝子産物の役割を調べるために、oep変異体に野生型由来の中軸中胚葉を移植して頭部を含む2次胚を誘導することを試みた。しかし、oep胚で移植により終脳まで含む頭部が誘導される頻度が非常に低く、従ってその背腹軸を調べることが現在までできていない。一方、終脳の背腹軸形成の研究過程で、前脳腹が領域の形成にFgf signalが関与していることが判明し研究を推進した。近年マウスなどでは、神経領域最前部で発現するFGFが終脳としての領域特異性の成立に重要であることが示された。ゼブラフィッシュにおいてもfgf8が神経領域最前部で発現しているが、その突然変異体であるacerebellar (ace)では終脳での異常はほとんど認められない。この為、ゼブラフィッシュにではFgfシグナル伝達の終脳形成における機能は不明であった。我々は、ゼブラフィッシュ胚においてFgfシグナルの伝達に必須なRas/MAPKカスケードが前脳最先端部で活性化されていることを燐酸化ERKに対する抗体染色で明らかになった。この領域は将来の前脳腹側を形成する領域であり、Fgf3とFgf8をともに発現している。次に、ドミナントネガティブ型に改変されたRas(Ras^<N17>)の過剰発現を行った結果、前脳腹側のマーカー、dlxの発現が抑制された。この表現型は、FGF受容体の特異的阻害剤(SU5402)の局所的投与によっても得られた。これらの結果は、Rasを介したFgfシグナルが終脳の腹側領域の確立に関与していることが示された。
