研究概要

1. 三叉神経脊髄路核および脊髄から中脳中心灰白質に直接投射するニューロンのなかにsubstance P 受容体(SPR)を発現するものがあるかどうかを調べた。実験動物はラット。方法は、Fluoro-gold(FG)を標識物質とする逆行性標識法とSPRの蛍光(Texas Red:TR)免疫組織化学を用いる二重標識法によった。
SPR様免疫活性(TR陽性)を示すニューロンのうち、中脳中心灰白質に注入されたFGによって逆行性に標識されるものは主として外側脊髄核、脊髄I層、および三叉神経脊髄路核尾側亜核I層に分布していた。また、さらに一部は脊髄V層・X層に分布していた。
2. SPないしCGRPを発現する三叉神経節ニューロンおよび脊髄後根神経節ニューロンがmu-opioid受容体(MOR)を発現するかどうかを調べた。
実験動物はラット。方法は、TRおよびdichlorotriazinyl aminofluorescein(DTAF)を用いる二重蛍光免疫組織化学、さらにimmuno-peroxidase法とimmuno-gold法の併用による電子顕微鏡的二重免疫組織化学によった。
三叉神経脊髄路核尾側亜核I層とII層、および脊髄I層とII層において、SP様ないしCGRP様免疫活性を示す軸索終末のなかにMOR様免疫活性を示すものが観察された。
