虚血性神経細胞死における新規ストレス蛋白ORP150の役割

研究概要

本年度は主に遺伝子工学的手法を用いて虚血性神経細胞死における 150 kDa Oxygen Regulated Protein 150(ORP150)の役割を明らかにすることを目的とし、以下の実験結果を得た。
(I)ORP150の発現機序の解析
ORP150は培養神経細胞において低酸素環境下において2-4時間と早期に一時的な比較的弱い誘導が認められた。一方アストロサイトにおいては6-12時間後より持続的な強い発現誘導が認められた。
(II)マウス中大脳動脈閉塞モデルにおけるORP150の発現の解析
免疫組織化学的解析により動脈閉塞3時間後にはORP150の発現誘導が虚血中心部の神経細胞に認められた。一方、虚血再潅流後3日目には虚血周辺部のアストロサイトにORP150の発現誘導が認められた。
(III)神経特異的にORP150を過剰発現するトランスジェニックマウスの作成
PDGF-b promoter下にORP150 cDNAを組み込んだtransgeneを発現するトランスジェニックマウスを作成し、ORP150の発現をNorthern blot,Western blot,免疫組織化学法にて確認した。大脳皮質、海馬、小脳において高い発現が認められた。
(II)ORP150トランスジェニックマウスにおける梗塞サイズの解析
マウス中大脳動脈閉塞後の梗塞サイズをTTC染色によりORP150トランスジェニックマウスと野生型マウス間で比較した。トランスジェニックマウスにおいて有意な梗塞サイズの減少が認められた。また、神経学的所見においても有意に改善が認められた。
