研究概要

新規神経ペプチドPACAPの含有ニューロン終末は視床下部の視索上核や室傍核に密に分布し、その神経終末が神経内分泌細胞にシナプスを形成しているのを我々は免疫電顕的に観察・報告している。免疫組織化学法とIn situハイブリダイゼーション法の組み合わせ法を用いて視索上核をしらべたところ、バソプレシンニューロンが特異的にこのレセプターを発現しているのが明らかになった。バソプレシンニューロンはノルアドレナリン性ニューロンの神経支配も受けており、ノルアドレナリンおよびPACAPはcAMP-プロテインキナーゼA系を介して細胞内のカルシウム濃度を上昇させることが単離神経分泌細胞を用いた実験で明らかになった(Shioda et al.,1998,1999)。特異的なカルシウムチャンネル阻害剤を用いて上記と同様の実験を行った結果、PACAP/ノルアドレナリンは共調して細胞膜のT-,L-型のカルシウムチャネルをリン酸化し、細胞内に大量のカルシウムを流入させ、最終的に分泌果粒の放出が起きることが明らかになった(Shioda et al.,1999)。
