誘導型一酸化窒素合成酵素の神経細胞特異的な発現に関わる細胞内シグナル伝達系の解明

研究概要	ヒト神経細胞でのiNOS遺伝子のサイトカイン刺激による発現誘導に関わる細胞内シグナル伝達系はほとんど解明されていない。我々は、ヒト神経芽腫NB-39-nu細胞でのiNOS発現誘導がTNF-α単独刺激では認められず、IFN-γ同時刺激にのみ認められる事を見出した。本研究では、サイトカイン刺激による神経特異的iNOS発現誘導に関わる細胞内シグナル伝達系の解明を行った。NB-39-nu細胞でのiNOS尭現誘導におけるIFN-γ役割は、従来のiNOS遺伝子発現調節領域に存在するエンハンサー領域IREやGASへの応答とは異なり、TNF-αおよびTNFレセプターII(TNF-RII)の発現誘導によるTNFシグナル伝達系の活性化を担っていることを明らかにした。事実、TNF-RIIcDNA導入NB-39-nu細胞(NB-39-nu/TNF-RII)ではTNF-α単独刺激によりiNOS発現誘導が認められ、この誘導はNF-κB活性化阻害剤PDTCにより完全に抑制された。iNOS遺伝子転写開始点近傍に存在するNF-κB配列を用いたゲルシフト法により、NB-39-nu/TNF-RIIのTNF-α激によりTNF-RIIを発現しないNB-39-nu細胞では検出されないNF-κB結合タンパク質の特異的活性化を認めた。このように、神経細胞でのiNOS発現誘導にはTNFおよびTNF-RIIシグナル伝達系を介するNF-κB活性化による新たなiNOS発現制御機構が存在する事を明らかにした。次年度は、ヒトiNOS発現調節領域を用いたプロモーター解析を通して、新たなiNOS発現誘導に関わるシスエレメントとその配列を認識する転写因子を同定し、神経細胞でのTNF-RIIを介する細胞内シグナル伝達系の解明を行う予定である。
