研究概要	研究代表者らは、Betzらが開発したシナプス小胞を標識する蛍光色素FM1-43をショウジョウバエ変異株に適用して、神経末端のboutonには、通常のexocytosis/endocytosisに関与しているシナプス小胞プール(exo/endocycling pool)と通常は関与していない小胞プール(reserve pool)が存在することを、報告した(Neuron 1998).今回は、各シナプス小胞プールのシナプス信号伝達における役割を明かにするため、神経刺激の頻度と各プールの関与について、実験を行なった。 1)低頻度刺激の時は、exo/endo cycling poolからのみ小胞が放出され、endocytosed小胞はexo/endo cycling poolにのみ取り込まれた.一方、高頻度刺激の時は、両方のプールから小胞が放出され、endocytosed小胞は両方のプールに取り込まれた。 2)adenylyl cyclaseおよびPKAの活性化薬、阻害薬の効果から、高頻度刺激によるreserve poolからexo/endo cycling poolへの小胞の移動にはcAMP/PKAカスケードが関与していることが明かになった。 3)学習能力に欠損のある変異株、dunceとrutabagaでは、reserve poolからexo/endo cycling poolへの小胞の移動が障害されていることが示され、シナプス信号伝達の可塑性に、シプナス小胞プールが極めて重要であることが明かになった。
