研究概要	1 センダイウイルスのマウス肺病原性を支配する遺伝子の同定センダイウイルスの毒力を規定するウイルス側の要因として、F蛋白のプロテアーゼによる活性化部位の構造、M蛋白による出芽方向の制御、およびその機構は不明であるがV蛋白のC端側、が報告されていたが、新たに新鮮分離株由来の強毒野生株(M11)と弱毒変異株(MVC11)との全塩基配列の比較から、C蛋白の170番目のPheからSerへの変異により、ウイルスの毒性がほとんど消失することを明らかにした。C蛋白の病原性への関わりを更に検討するため、4種類のC蛋白(C',C,Y1,Y2)を様々に欠損した変異ウイルスをリバースジェネティクス法で作製したところ、C'とC、およびCのみを欠損した変異株で、マウス病原性が著しく減弱していた。2 C蛋白変異ウイルスの弱毒化機構これらのC蛋白変異弱毒化ウイルスは、いずれもアポトーシスおよびネクローシスを引き起こし、感染細胞を死滅させた。その結果、ウイルスの増殖は中断され、このことが病原性の低下をもたらすものと推察された。一方、強毒株によるアポトーシスおよびネクコーシスの誘導は軽微で、長期間にわたりウイルスを産生し続けた。カスパーゼ阻害剤存在下でも、弱毒株感染細胞は強い細胞変性を示し、速やかに死滅した。この条件では、アポトーシスの誘導は抑制されていたが、ネタローシスは抑制されておらず、弱毒株による細胞死にはネクローシスの役割が大きいと考えられた。 3 組み換えキメラウイルスの増殖能センダイウイルスZ株と新鮮分離株のキメラウイルスをワクチン株として用いるため、遺伝子の組み換えがウイルス増殖に及ぼす影響を調べた。Z株のP遺伝子の一部を新鮮分離株のものと置換した場合、培養細胞での増殖能が低下する傾向が認められ、今後詳細に検討する必要があるものと考えられた。
