研究概要

1.Rhodovulum sulfidophilumの光合成反応中心遺伝子欠損株の作成
R.sulfidophilumの光合成中心反応遺伝子の一部を薬剤耐性遺伝子に組み替えた。変異株には光合成器官合成能が見られず、反応中心遺伝子も欠失していることが確認された。
2.R.sulfidophilumにおける光合成反応中心遺伝子相補実験
(1)R.sulfidophilumの反応中心遺伝子を挿入した広宿主域プラスミドを上記R.sulfidophilum光合成反応中心欠損株に導入したところ、光合成器官合成能が回復した。
(2)しかし、Acidiphilium rubrumの光合成反応中心遺伝子を挿入した広宿主域プラスミドを導入したところ、反応中心遺伝子の発現は見られなかった。
3.R.sulfidophilum光合成反応中心遺伝子発現のためのプロモーター部位の解析
lacZレポーター遺伝子を用いたmRNAの解析により、R.sulfidophilumの反応中心遺伝子の発現には上流の3つのプロモーターが働いていることが明らかになった。
A.rubrumの反応中心遺伝子がR.sulfidophilum中で発現しない原因はプロモーター配列の違いによると考えられる。3で明らかとなったR.sulfidophilumのプロモーター配列をA.rubrum反応中心遺伝子に導入し形質転換を試みる必要がある。また、2(1)により、R.sulfidophilum反応中心遺伝子に対する形質転換系が確立した。亜鉛型バクテリオクロロフィルを持つA.rubrum反応中心に特異的なアミノ酸をR.sulfidophilum反応中心に導入することにより、クロロフィルと色素タンパク質間の相互作用の研究を展開する予定である。
