神経細胞の酸化ストレスとその防御因子に関する研究

研究概要	神経変性のメカニズムとその防御因子の探索を行った。代表的神経変性疾患であるアルツハイマー病(AD)では、特徴的の1つとして脳内でアミロイドβ蛋白(Aβ)沈着が認められ、病理学的に老人斑として知られている。Aβの沈着はと神経変性の原因と考えられるとともに、逆に神経防護反応の1つとも考えられる。平成10〜11年度の研究により、我々はADの病因に密接に関連すると考えられているAβ42が、神経細胞のアポトーシスプロセスを誘導することで細胞内で増加・沈着することを見出した(業績論文#1)。一方,一次的な核DNA傷害を伴わないネクローシスプロセスにおいては、このようなAβ42の増加・沈着は認めなかった。このAβ42沈着は細胞死よりも早期に出現し、核DNA傷害などに対する防獲因子として働いている可能性が考えられた。また、細胞内Aβ42沈着は筋炎組織の筋衛星細胞内にも見られ、神経細胞以外の細胞でも、何らかの基礎的な細胞代謝調節機能を有している可能性を明らかにした(投稿中)。さらに現在、Aβは正常細胞内では24kDの可溶性複合体蛋白として小胞体やゴルジに存在し、神経細胞への分化や細胞周期の停止に伴い核内に移行することを見出している。培養細胞にAβ42を発現させるとp53プロモーターの活性を上昇させ、その結果細胞周期の停止や核DNA傷害の防護に関与している可能性がある。現在、このような分子レベルでの細胞内Aβの機能や病的な異常を解析中であり、このようなメカニズムの解明がADにおける神経変性の解明や治療法の開発に貢献すると考えられる。
