アルツハイマー病発症におけるプレセニリンの意義について

研究概要	家族性アルツハイマーの発症機構を知るため、プレセニリン遺伝子の変異がアルツハイマー脳に沈着するアミロイドβペプチド(Aβ)の分泌に与える諸要因を調べた。N141lアルツハイマー変異を有するプレセニリン2のカルボキシ末端側に様々な欠損や修飾を加え、培養細胞に発現し、分泌されるAβを測定すると同時に、プレセニリン蛋白のプロセシングと安定性を検討した。N141l変異をもつプレセニリン2のカルボキシ末端3ないし7残基を欠損させると、全長分子のもつAβ42分泌増大効果は完全に抑制された。最C末端に位置するlle448を様々なアミノ酸に置換すると、特に疎水性の低いアルギニンに変化させた場合に完全なAβ42の抑制が見られた。C末端へのヒスチジン等のアミノ酸を付加すると、同様にAβ42は抑制された。このときAP42増強効果はプレセニリン2蛋白の安定化の程度に比例して生じていることが判明した。このことから、安定化を受けた断片型プレセニリンが活性型分子であること、プレセニリンの最C末端に安定化とAβ42効果にかかわる重要な機能ドメインが存在することが明らかとなった。今後このドメインがプレセニリン分子の全体に与える影響や、この部位に結合する蛋白について検討を行う。またPS1のN末端とPS2のC末端を持つキメラ分子も、PS1のN146L変異を加えるとAβ42増大効果を発揮し、しかもPS1N末端とPS2C末端のヘテロ2量体を形成して安定化されることが判明した。
