内因性の学習障害誘発因子の単離同定とその生理機能・発現制御機構の解明

研究概要

学習障害モデル動物として確立しつつある両側総頚動脈永久結紮(2VO)ラットの結紮にともなって脳内で生ずる分子的変化を遺伝学的手法を用いて検討した。明らかな学習障害を生ずる術後4カ月のラット脳よりDifferential Display法によって、発現量が増減する因子を数種単離し、その配列を決定した。それぞれの得られた配列が、はじめの単離産物と一致することを制限酵素等により確認した後、得られた因子のうち、2VOにより発現量の増大するvof-16(仮称、遺伝子バンク登録No.AB006881)について、さらに詳細に調べた。PCR法に基づく定量から、vof-16は海馬、大脳皮質に多く発現しているこが認められた。in situ hybridization法によりその分布をさらに詳細に検討したところ、やはり海馬に顕著に多いこと、皮質全体に広がっていることが確認できたが、皮質ではpiriform cortexに特に多いこと、その他の部位ではtenia tectaに顕著に多く、また大動脈血管周囲の細胞にも多く発現していることが認められた。海馬におけるvof-16発現量は結紮によって2〜4倍程度増大すること、ならびにその発現量の増大と学習障害の程度に相関性があるかもしれない点が、定量的PCR法により示唆された。現在その詳細ならびにvof-16の全長についての検討を続けている。また、学習障害の原因因子の検索を目的として、結紮初期(4日後)に発現変化する因子についても、同様の手法により検討を行ない、いくつかの因子を単離し、その詳細を調べている。
