老化モデルとしてのミトコンドリアDNAノックアウトマウスの樹立と応用

研究概要	高齢化社会を迎た現在、老化と糖尿病は重大な社会問題となりつつある。ごく最近、ミトコンドリア遺伝子疾患の原因となるミトコンドリアDNA(mtDNA)の突然変異と全く同じ突然変異が、実際にはごく少量ではあるがほとんど全ての老化したヒトの組織で発見されたばかりでなく、かなり多くの糖尿病患者やアルツハイマー病患者の組織のmtDNAにも存在することが相次いで報告された。これによってmtDNAの突然変異とミトコンドリア遺伝子疾患、老化、神経変性疾患、糖尿病との因果関係はにわかに大きな注目を集めるようになった。つまりmtDNAの突然変異はミトコンドリア遺伝子疾患という特殊な病気に限定されず、極めて身近なもので健常者一人一人の体の中にも少量ながら存在し、われわれの健康に重大な影響を与えている可能性が出てきたのである。しかしmtDNAの突然変異が本当に老化などの原因であることが直接証明されたわけではない。そこで本プロジェクトでは、我々が樹立したヒトmtDNA欠損細胞を用いたmtDNA導入系を活用することにより、ミトコンドリア遺伝子疾患を引き起こしたものと全く同じmtDNA突然変異が、老化や糖尿病などの多様な症状を引き起こすかどうかを調べ、両者の因果関係を直接立証することを試みている。一方、ごく最近樹立したマウスmtDNA損細胞を利用することにより、ヒトと同じmtDNA突然変異を持つマウスの作製に成功し、この論文は、Nature Geneticsの2000年10月号に掲載され、マスコミの注目も集めた。これらを病態モデル動物は今後更に、ミトコンドリア遺伝子疾患、老化、糖尿病に限らず、mtDNA突然変異が起こす多様な臨床症状に対する有効な治療方法の手かがりを得上で有効に活用できるものと期待されている。なお、このマウス作製法に関しては、筑波リエゾン研究所を通して特許出願を完了している。
