Th1/Th2バランス制御因子の遺伝子解析とその免疫疾患との関連性

研究概要

本年度は、マウス免疫制御の中枢を担うThl/Th2バランスの制御因子に関する研究、およびThl/Th2バランスを支配する遺伝子のクローモゾームマッピングについて検討を行った。その結果、以下のような事実を明確にさせることができた。
1) Thl免疫を活性化できるIL-12の生体内投与によって、抗腫瘍エフェクター細胞がリンパ節に移動する、Habbitat segregationの現象を見いだした。
2) IL-12の生体内投与によって、免疫能の活性化のみならず、脾細胞における髄外造血が促進されることを明らかにした。
2) 拘束ストレスによりナチュラルキラー、IFN-γは著しく減少するが、IL-4産生はそれほど低下しないことより、ストレスにより、Thl/Th2バランスはTh2偏向することをはじめて明確にした。
3) ヒトアトピー性皮膚炎患者末梢血においては、Thl免疫の低下が著しく、IFN-γ産生能が低下していることを明らかにした。また、抗原特異的なメモリー細胞の頻度も低下していることも見いだした。
4) BALB/cはIL-4やIL-10を遺伝的に高く産生しやすく、Th2に偏向しやすいマウス系統と考えられる。そこで、IL-4の高産生を支配する遺伝子のクロモゾームマッピングを行った結果、マウスクロモゾーム7番と13番に強い連鎖が確認された。特にマウス7番染色体はヒト16q12とシンテニーがあり、その近傍にはIL-4レセプターが存在することも明らかにされた。
以上の成果をもとに、さらにThl/Th2バランス制御因子を追求して、人為的免疫制御の免疫病治療への応用を計りたいと考えている。
