Th1/Th2バランス制御因子の遺伝子解析とその免疫疾患との関連性

研究概要	免疫機構は神経、内分泌系と同様に生体の恒常性維持に重要なシステムであると言える。免疫バランスが正常に保たれ、自己および非自己抗原の識別が正しく行われている場合には免疫病発症の危険性はないが、一端、その機構に破綻が生ずると、多くの免疫病発症の原因となりえる。最近の免疫学の進歩によって、免疫バランスの制御が2種類のヘルパーT細胞サブセット(Th1,Th2)を軸として行われていることが判明してきており、Th1/Th2バランス制御と免疫疾患に関わる研究が、免疫疾患に対する新しい診断、治療法開発に重要な課題であると考えられている。そこで、本研究においては、Th1/Th2バランスの制御とその免疫病発症における意義に関して検討をおこなった。また、Th1/Th2バランスの遺伝的支配因子を追求することによって、遺伝性免疫疾患に関連性があると思われる因子を遺伝子レベルで解明することを試みた。その結果、以下の点を明確にすることができた。 1)各種免疫疾患におけるTh1,Th2細胞の役割を、癌、アレルギー、肝障害の新しい疾患モデルを作製することによって明確にさせた。同一のナイーブThから誘導されたTh1,Th2細胞が全く異なった機能を有して、違った病態発症に関わっていることが示された。 2)ヒトアトピー性皮膚炎患者におけるTh1/Th2バランスの異常を明らかにした。すなわち、アトピー患者においては、Th1免疫の調節に重要なIFN-gの産生能が著しく低下し、結核菌等に対する細胞性免疫記憶能も低下していた。 3)マウスIL-4産生能を指標として、Th1/Th2バランス制御に関連性を示す遺伝子を染色体上にマッピングすることができ、候補遺伝子の一つにIL-4レセプターα遺伝子が存在することを明らかにした。
