遺伝子の分節構造に着目したゲノム遺伝子の自然史科学的研究

研究概要	ゲノム解析として、ヒト組織適合性抗原遺伝子領域の塩基配列決定をその進化的解析を行い、その領域が大規模な複製やSINEやLINEなどの反復配列の挿入や欠失などの進化的要因によってダイナミックに変化していることを明らかにした。また、この領域の中には未知の遺伝子が数個発現している可能性があることを示した。つまり、ゲノムはこのような大規模変化と突然変異などの小規模変化を綯い交ぜにしながも、時々刻々と進化しているのである。タンパク質のレベルでは、特にperiplasmicタンパク質群の高次構造の進化的解析を行い、高次構造の進化的変異がアミノ酸配列から推定される系統進化に一致することを示した。さらに、このタンパク質群の進化経路に沿って、高次構造がどのように変異を遂げてきたか詳細に推定した。それによると、このタンパク質群を構造的に2つ分ける大規模な構造変化は進化過程でただ一回起こり、その後微少変化が続き現在のタンパク質群に進化したということになる。また、系統進化研究面では、ミトコンドリア全領域の塩基配列情報にもとづいて、多種の脊椎動物の系統関係を解析した。その結果、部分領域から得られた系統関係とは違った、しかもより信頼できる脊椎動物の系統進化の描像が明らかになった。これらの研究成果は、生物の自然史を遺伝子あるいはタンパク質レベルで解析する道筋を拓き、より定量的な結果にもとづいて議論を進めることを可能にしたといえる。これらの成果は関連分野の国際-流誌に発表されたか印刷中である。
