巨大ベシクルのレーザー分裂・融合・構造形成制御と二分子膜構造

研究概要	Didodecyldimethylammonium bromide(DDAB)は水中におて数マイクロメートルから数十マイクロメートルのジャイアントベシクルを形成する。一方、集光したレーザー光が物質により散乱されるときに発生する放射圧を利用することによりマイクロメートルの微粒子から比較的に大きな分子や分子集合系を捕捉することが可能である。本研究においては、DDABベシクルをレーザー光により分裂、構造制御するとともに、顕微分光測定を通してベシクルを構成する二分子膜構造の再構成過程をその場測定することを目的として研究を行った。 1)単一ベシクル形態ののレーザー制御 : ベシクルをレーザー捕捉するため、2本のレーザー光で2箇所を独立に捕捉することのできるダブルビーム走査型レーザーマニピュレーションシステムを構築した。これにより、約10マイクロメートル程度の単一ベシクルの両端をレーザー捕捉し、一方の集光レーザー光をXY平面内で走査することによりベシクルを球形から楕円形へ変形させること、更には分裂、2つのベシクルの融合に成功した。 2)単一ベシクルの顕微分光 : DDABベシクルをFluorescein等の蛍光色素で染色して、単一ベシクル毎に顕微分光測定を行った。その結果、蛍光色素の極大波長と寿命はベシクルのサイズに依存することが見い出された。 3)ベシクルの膜構造変化の電気科学制御 : 分子内に電気化学活性なフェロセン部位を有するDDAB誘導体を合成した。DDABとの混合ベシクルをマイクロ電極に接触させて電気分解することにより、単一ベシクルの消滅を電気科学的に制御可能であることを示した。先に述べたレーザーによるベシクルの分裂・変形操作と蛍光分光法を組み合わせることにより、二分子膜の再構成過程の情報が得られるものと期待される。
