慢性ウイルス肝炎における肝癌の成因:フリーランジカルと一酸化窒素代謝物による感染性発癌機構

研究概要

感染・炎症において過剰に生成するスーパーオキサイドやNO由来のパーオキシナイトライト(ONOO^-)は、タンパク質や遺伝子(塩基)のニトロ化をもたらし、生体内で酸化ストレスや変異原性を発現していることが示唆されている。そこで本年度は、生体内二トロ化反応の解析のためのマーカーとしてのニトロチロシンの高感度検出法の確立と、ウイルス遺伝子変異に対する反応性NO酸化物(特にONOO^-)の影響を検討した。
その結果、ニトロチロシンの検出については、多電極型電気化学検出器を接続したHPLC法(HPLC-array-EC法)により (1)ニトロチロシンは、絶対量で0.1pmolと非常に感度よく検出することができた。(2)タンパク質中に生じたニトロチロシンは、酸加水分解あるいはタンパク分解酵素で処理することで検出可能であった。(3)これまで免疫組織化学的にニトロチロシンの生成を確認しているインフルエンザ感染マウス肺組織中のニトロチロシン量を測定したところ、チロシン100残基あたり0.03残基がニトロ化されているという結果が得られた。これらの結果はHPLC-array-EC法がニトロチロシンの定量法として優れていること、および、免疫組織化学法と組み合わせることで生体内ニトロ化を定量的に、より詳細に検討できることを示唆している。
つづいて反応性NO酸化物のウイルス遺伝子変異について、変異のマーカーとしてgreen fluorescent protein(GFP)を組み込んだセンダイウイルス(GPF/SeV)を用いて検討した。その結果、invitroの系において、GFP/SeVを生理的濃度のONOO^-(0.8μm)で処理すると、ウイルスの感染性が損なわれない条件下で、ウイルスゲノムに組み込まれたGFP遺伝子が変異した株が、0.5%-1.0%の頻度で出現した。また、ONOO^-がウイルスに対して99.98%の致死率を示す反応条件下では、100%の変異率が得られた。以上の知見は、感染・炎症において過剰に生成するNOやONOO^-がウイルスゲノムの変異を促進する因子として働く可能性を示している。
