免疫組織に発現するFormin関連遺伝子frlと免疫臓器の発生と構築の制御

研究概要	我々が単離したfrl遺伝子は、リンパ組織(リンパ節、脾臓、骨髄、胸腺)に発現するFormin関連遺伝子である。そのcDNAは約3.6kbでコードする蛋白は1064アミノ酸から構成され、SDS-PAGEにおける分子量は約180kDaであり・Frlタンパクは、プロリンに富む領城FH1とformin familyで保存されているFH2領域を有する新規のタンパクである。frl遺伝子はマウス染色体第11番17q21に位置し、しかも,Alyマウス(リンパ節、パイエル板を欠損し、脾臓構築に異常があり濾胞構造をもたない先天性免疫不全症マウス)の遺伝子の近傍にマップされた。遺伝子構造解析の結果、frl遺伝子とaly遺伝子は10kb以内の距離で隣り合っていることがわかった。Alyマウスの胸腺、脾臓ではfrlタンパクの発現の著明な低下が見られる。frlゲノム遺伝子を単離し、ターゲティングベクターを構築して相同組換え法により、Frl欠損マウスを作成した。Frl欠損マウスにおけるリンパ組織の異常は見出されなかった。Frl欠損マウスをT依存性抗原で免役し、その一次反応、二次反応を調べたところ、二次反応においてIgG2a産生の異常亢進、IL2、IFNγの産生亢進が見られた。このことはThlヘルパーT細胞の生成、メモリーT細胞(又はB細胞)の生成の場においてFrlタンパクが何らかの機能をしていることを示唆している。今後、Frlタンパクの機能と抗原免疫後の免疫組織の構築、免疫学的記憶の成立との関連について研究を進める。
