転写因子の生体内分子構成による新規抗炎症療法の開発に関する研究

研究概要

本研究はグルココルチコイド受容体をモデルに用いて、転写因子のドメイン特異的機能を時間特異的に、しかも低分子量の薬剤を用いて発現させる方法論を樹立し、最終的に転写因子の機能制御を標的とした炎症性疾患の新たな治療法を確立することを目的としている。そのためにはモデルとした転写因子の各機能ドメインを明らかにし、その制御機構を究明することが最重点課題である。
かかる目的を達成するにあたり、まず、分子生物学的方法を駆使してグルココルチコイド受容体の各ドメインの機能を詳細に究明した。2年間で、とくに、DNA結合ドメイン、リガンド結合ドメインの機能解析が進展し、リガンド結合、核移行、転写共役因子との相互作用に重要なドメインを同定できた。とくに、リガンド結合領域の機能を多様に制御する薬理学的方法論を確立できたことは大いに意義がある。すなわち、C末端領域の数十個のアミノ酸がりガンドによる核への移行ならびに転写共役因子との相互作用の制御にきわめて重要な働きをしていることが明確になった。同時に、かかる領域を合成グルココルチコイドをはじめとした薬剤が多様に、あるいは、各作用選択的に制御することを見い出した。これらの成果を得る過程で構築したプラスミドは二量体形成実験にそのまま使用できた。トランスジェニックマウスは当初の予定より施設の都合で遅延しているが、1年以内に初期の目標を達成可能と考える。
