アポトーシスの誘導と死細胞除去におけるオキシステロールの役割

研究概要	オキシステロールはコレステロール酸化物の総称であり、細胞障害作用等の多様な生体内機能を有する。しかし、その詳細なメカニズムは不明である。プラークの安定性が急性心筋梗塞の発症素因として重要であることが明らかにされてきた。動脈硬化モデル動物のアテローム性動脈硬化病変におけるアポトーシス細胞の分布をterminal deoxynucleotidyl transferase-mediateddUTP nick end labeling(TUNEL)法を用いて検討した。その結果、TUNEL陽性細胞は、泡沫細胞のみが観察される初期病変に多数確認されることを見い出した。これらの細胞は、マクロファージか血管平滑筋に由来することを免疫組織染色によって確認した。細胞の泡沫化がアポトーシスに先行し、泡沫化が直接あるいは間接的に細胞死を誘導していると考えられた。一方、単球マクロファージ系の細胞株であるP388Dl細胞を用いた検討では、LDLやアセチルLDLと細胞をインキユベートしても泡沫化のみ観察されて、細胞死は観察されないが、酸化しLDLとのインキュベートでは、細胞の泡沫化とともに細胞死が観察された。その場合、DNAの断片化やアポトーシス体が観察されることから、ネクローシスではなくアポトーシスが細胞死の正体と考えられた。酸化しLDLのどの成分がアポトーシスを誘導したかについて、酸化しLDLの成分について、細胞死の誘導能を検討したところ、25ヒドロキシコレステロールや7ケトコレステロールなどのオキシステロールに最も強いアポトーシス誘導活性が存在することが明かとなった。オキシステロールによるアポトーシス誘導作用は、Bcl-2の発現仁よって部分的に抑制され、その経路はCaspase-3(CPP32)の抑制を介していることを明かにした。
