モデルマウスを用いた無精子症の遺伝子治療と精子生成過程の分子レベルでの解析

研究概要

ベイシジン(Bsg)は免疫グロブリンスーパーファミリーに属する膜糖タンパクである。Bsg(-/-)胎仔の多くは着床の前後で失われた。一方、成体に達すると両性とも不妊であった。雄性不妊の原因は無精子症で、第一次減数分裂前期で分化が止まっており、これは、野生型マウスの精巣でのBsgの発現とよく一致した。雌性不妊については、卵成熟と着床の二段階で異常のあることを明らかにした。また、記憶学習、匂いに対する反応性等の神経学的異常をきたし、野生型マウスの中枢神経での発現様式とあわせて考えて、機能的神経ネットワーク形成にBsgが重要であることが示唆された。また、Bsgが細胞膜上でホモ2量体を形成し、これにはN末側の免疫グロブリンドメインが重要であることを明らかにした。
Bsg発現ベクターを電気穿孔法を用いて、Bsg(-/-)雄の精彩管内に導入すると、導入後1週では、生殖細胞の発現がみられるが、4週では主な発現の場所がセルトリ細胞であることが判明した。現在、発現効率を上昇させるとともに、Bsg導入による生殖細胞の分化の詳細な解析を進めている。
