胎盤でのアポトーシス抑制遺伝子bcl-2発現の意義の解析とその周産期病態への関与

研究概要

ヒト胎盤絨毛組織において、幹細胞である細胞性栄養膜細胞から分化し、母体脱落膜深部へ侵入する細胞集団は絨毛外栄養膜細胞と称される。この絨毛外栄養膜細胞の脱落膜深部への侵入と、生体のホメオスターシス維持の上で細胞が本質的に備えた自己消去機構であるapoptosisとの関わりを調べるため、胎盤組織切片上で、DNA断片化シグナルならびにapoptosis関連因子であるbcl-2蛋白、Fas/Fas ligandの発現態度について検討した。DNA断片化シグナルの発現態度はin situ DNA 3′-end labeling法によって調べ、絨毛外栄養膜細胞の同定はhPLとcytokeratinの免疫染色により行った。
bcl-2蛋白は絨毛外栄養膜細胞に局在し、その発現レベルは脱落膜組織の浅部で強く、深部で減弱することを認めた。一方、FasとFas ligandの発現は脱落膜組織の浅部で弱く、深部へ進むにつれて増強した。正常末期胎盤の絨毛外栄養膜細胞には散在的なDNA断片化シグナルの出現をみたが、DNA断片化シグナル陽性率は脱落膜組織の浅部に比較し、脱落膜組織の深部で有意に高かった。正常末期胎盤の絨毛外栄養膜細胞におけるbcl-2蛋白、Fas/Fas ligandの局在態度ならびにDNA断片化シグナル発現態度より、絨毛外栄養膜細胞にはbcl-2によるapoptosis抑制機構が存在し、また脱落膜深部に到達した絨毛外栄養膜細胞ではFas/Fasligand発現を介してapoptosisが起ることが示唆された。
