エネルギー損失分光電子顕微鏡を用いた組織細胞化学による石灰化研究の新しい展開

研究概要

本年度は、電子エネルギー損失分光電子顕微鏡(EELS)の運用に改良を加え、今までなし得なかったÅレベルの元素の局在解析法と高分解免疫電顕法を開発し、生物学的石灰化におけるコラーゲン性石灰化の機序を解明することを目的として、EELSとイメージングプレートとを組み合わせた分析電顕システムを用いてコラーゲン性石灰化機構を解析した。
実験試料には、胎生期ラット頭頂骨を液体ヘリウムで急速凍結し、さらに凍結置換を行ったものを用いた。この試料を超薄切して、EELSで観察し、微細形態の検索や、元素分析、オステオネクチンの免疫電顕等を行った。
胎生期頭蓋骨では、基質小胞内での結晶析出から、石灰化の形成、コラーゲン性石灰化による広範な石灰化基質の確立にいたる一連の過程が観察できる。石灰化球では石灰化結晶の集積が観察されるが、結晶表面には、酢酸ウラン・クエン酸鉛の電子染色で染色される幅約1nmの結晶鞘が存在しており、その部位には、Nの明らかな集積は見られなかったことから、結晶鞘は比蛋白性成分から構成されていることが示唆された。このように、免疫電顕では物質の局在同定は困難である微細な構造においても、この電顕を用いた分析では、組成に関する情報がえられることが明らかとなった。
さらに、この電顕を用いた高分解能免疫電顕では、石灰化球とコラーゲン細線維の境界部にオステオネクチンが集積することが明らかとなり、オステオネクチンがコラーゲン性石灰化に重要な役割を果たすことが示唆された。
