軟骨細胞分化とイノシトールリン酸代謝に関する研究

研究概要	イノシトールポリリン酸ホスファターゼ(MIPP)遺伝子が、ニワトリの増殖軟骨細胞層/肥大軟骨細胞層の境界に特異的に発現する遺伝子Band17と極めて類似していた。本研究では、両遺伝子の類似性および軟骨細胞分化におけるイノシトールリン酸代謝系の係わりを明らかにすることを目的とした。6日齢ラット脛骨骨端成長軟骨を用いたinsituhybridization法により、MIPP遺伝子が増殖細胞層から肥大細胞層にかけて強く発現していることを明らかにした。これはBand17遺伝子で示されていた結果と同様であり、両遺伝子の類似性を示唆するものであった。軟骨細胞分化のモデルとして選択したAIDC5細胞をインスリン存在下で培養後、0、2、10、17、21、28、34日の各段階でRNAを調製した。ノーザンブロッティング法によりI型・II型・X型コラーゲン遺伝子発現を調べATDC5細胞の軟骨細胞としての分化段階を推定した。II型コラーゲン遺伝子発現の上昇がみられるインスリン刺激後10日目から、X型コラーゲン遺伝子発現の上昇がみられる17〜21日目にかけてATDC5細胞が軟骨細胞へと分化し、骨端成長軟骨での増殖細胞層/肥大細胞層に相当する段階にあると考えられる。MIPP遺伝子の発現を調べたところ、インスリン刺激前でも発現は認められるものの、刺激後2〜10日にかけて上昇し、その後も刺激前に比較すると高い発現を維持していることが分かった。したがって、モデル細胞株における細胞分化においてもMIPP遺伝子は増殖細胞層/肥大細胞層にかけて発現するものと思われた。軟骨細胞分化過程においてイノシトールリン酸代謝系を介した細胞内情報伝達経路が存在することが予測される。
