口腔扁平上皮癌細胞におけるnm23遺伝子の分化誘導抑制効果とその制御

研究概要

我々の教室で分離・樹立したヒト口腔扁平上皮癌細胞BHYは、ヌードマウスに移植すると、高度分化型扁平上皮癌を形成する。一方、ヒト口腔扁平上皮癌細胞HNは未分化型扁平上皮癌細胞を形成する。1.HN細胞では、nm23-H1及びnm23-H2遺伝子とも、mRNAが高度に発現していることを、一方BHY細胞ではそれぞれの遺伝子の発現量の低いことを、Northern blotting及び半定量RT-RCR法により明らかにした。更に、タンパクレベルでも同様の所見をimmunoblotting及び免疫染色法(ABC)により検出した。2.BHY,HN細胞にnm23-H1あるいはnm23-H2遺伝子に対するアンチセンス・オリゴヌクレオチドを導入したBHY,HN細胞において、TPAに応答してインボルクリン、フラゲリンあるいはサイトケラチンK1遺伝子が高度に発現してくることをABC法、蛍光抗体法あるいはNorthern blottingにより検出した。すなわち、nm23-H2遺伝子は分化抑制機能を保有していることを示唆する実験結果を得た。3.nm23-H2cDNAをSRαプロモーターにアンチセンス・オリエントに連結した発現ベクターをBHY,HN細胞にリポフェクチン法により導入した。このtransfectantsをTPAにてin vitroで処理した後、ヌードマウスに移植して、腫瘍増殖と腫瘍分化度につき検索した。その結果、有意な腫瘍の増殖抑制とkeratinocyte differentiationの出現を検出した。
