慢性脳虚血ラットの記憶障害に関与する内因性物質の分子生物学的研究

研究概要	両側総頸動脈永久結紮(2VO)ラットでは、記憶学習障害が徐々に進行し、結紮後2〜4ヵ月目に顕著になることを報告してきた。そこで学習障害発症前の脳慢性虚血初期の段階での生理的変化を明らかにし、それを制御することによる痴呆の治療薬・予防薬の開発をめざした研究をこの「萌芽的研究」によって行なった。昨年度は2VO処置後1〜7日間の脳組織変化を時間を追って検討し、白質の粗鬆化、海馬神経細胞の萎縮、樹状突起の消失とアストロサイトの発現等を報告した。本年度、これらの機序をさらに分子的に明らかにするために、2VO4日後の脳中で発現量の変化する遺伝子のクローニングをDifferential Display法により行なった。その結果11個のmRNA発現量の増大する因子を単離し、その全ての配列を決定した。得られた11種の因子のsense鎖を決定した後、定量的PCR法によりその発現量変化を、2VO処置群と対照群との間で詳細に検討した。発現量増大の程度が最も大きかったvof-21(単離配列は415塩基)について、その全長のクローニングを行なう目的で、5'-race,3'-race法を行なった。その結果、3'側ではこれ以上の配列は得られなかったが、5'側の266塩基の新たな配列が明らかとなった。明らかにした計681塩基は新規の配列であったが、種々の検討から、vof-21は5'側にさらに配列を有するものと考えられた。また、通常内部標準として用いられるGAPDHの発現量も2VO処置4日後に増大している事が認められたため、本研究では内部標準はβ-actinを用いた。現在、vof-21の全長についてさらに検討を加え、発現量の時間的変化や、発現部位についての詳細な知見を得ることにより、この因子の機能の解明を行なっている。また、咋年報告した、THAやGST-21のvof-21発現への影響について解明するとともに、その他の10個の因子についての検討も行なう予定である。
