紫外線によるDNA光生成物シトシン水化体の生物作用と修復

研究概要	太陽光線に含まれる紫外線は、突然変異、致死、発がんや老化を引き起こす。その原因は、紫外線によるDNA光生成物であるピリミジン2量体及び6-4付加体と考えられている。紫外線はその他に、ピリミジン2量体の5〜10%程度を占めるシトシン水化体を作る。シトシン水化体は、ピリミジン2量体を修復する除去修復系及び、チミングリコール等の修復酵素、エンドヌクレアーゼIII/VIII、の2つのシステムによって修復されている。シトシン水化体の生物作用を明らかにするために、除去修復のないuvr欠損及びエンドヌクレアーゼIII/VIIIの欠損したnth nei変異株からuvrA nth nei3重変異株を作成した。この大腸菌は、uvrA株より紫外線に感受性となり、nth nei株より過酸化水素に感受性となった。紫外線照射したsupF遺伝子を持つプラスミドを、ピリミジン2量体及び6-4付加体の両方を修復する光回復酵素で処理し、突然変異を見たところ、G:C→T:A及びG:C→C:Gトランスバージョン変異が増加した。ピリミジン2量体及び6-4付加体以外のDNA損傷がトランスバージョン変異誘発に関わることが明らかになったが、この原因がシトシン水化体であるかどうかは不明である。光回復処理したプラスミドをin vitroで更にエンドヌクレアーゼIII処理してシトシン水化体を取り除き、そこでの変異を観察する必要がある。今回更に、uvrA nth nei3重変異株が、過酸化水素にも感受性であることが明かとなった。紫外線での感受性の結果と考えて、紫外線照射で過酸化水素が生じている可能性もあり、今後の検討課題である。尚、本研究の成果は、放射線影響学会第40回大会で発表した。
