バクテリアが生成するアポトーシス抑制因子の同定とそのメカニズム

研究概要

われわれの研究室では細胞内情報伝達機構におけるチロシンキナーゼSykの役割について解析してきた。とくに最近、酸化的および種々のストレスによるBリンパ球のアポトーシス制御機構においてチロシンキナーゼSykが重要な役割をしていることを証明してきた。その研究過程において偶然、放線菌などのバクテリアの分泌物がある種のアポトーシスを抑制する現象を見いだした。われわれはこれらのバクテリアがある種の活性酸素やNOに抵抗性を誘導する物質を生成しているのではないかと考え、スクリーニングを試みたところ、ある種の細菌において、カタラーゼなどの消去酵素そのものではなく、消去酵素系の活性化を誘導する蛋白質を分泌していることが示唆された。今回、この因子の同定を試みた結果、ある酵素がその原因物質として同定された。この酵素は様々なアポトーシスを抑制する効果をもつことが明らかとなったが、それ以外にも興味深い作用がいくつか認められた。この酵素はほ乳類にも存在することが知られているが、現在のところその機能はほとんど不明である。現在、ほ乳類の酵素を精製分離中である。今後、さらに噛乳類の細胞系におけるアポトーシス抑制活性をさまざまなアポトーシス誘導刺激(FAS抗原、紫外線照射、セラミドなど)にて詳細に解析し、その分子メカニズムを解明したい。残念ながら、未だ研究が予備段階なので、もう少しはっきりとした結果が得られて後、具体的な実験結果およびこの酵素名を明らかにする予定である。
