研究概要

「類人猿の進化と人類の成立」という課題で、1)社会・生態、2)形態・古生物、3)認知・脳科学、4)分子・遺伝の4班構成により、この5年間研究を進めてきた。これは「霊長類の進化を明らかにし人類の起源を考察する」とする霊長類学の根本的な課題であると考えている。以下最終年度の成果に加え、この5年間の成果について述べる。
社会・生態について。コンゴ民主共和国(旧ザイール)ワンバ森林のピグミーチンパンジー(ボノボ)研究は日本の研究グループが世界に先駆けて始め、数多くの発見があったが、内戦によりここ10年程中断していた。その研究再開が期待される報告が分担者・古市によりなされた。かつて識別されていた対象個体のうち、一頭を除き現在も生存していることが明らかになったのである。分担者・西田らにより長年おこなわれているチンパンジー研究拠点・マハレ(タンザニア)での詳細な行動研究も、毛づくろい行動研究などに大きな進展があった。また分担者・山極によって「類人猿比較から人間性について何がわかるのか」という深い考察がなされた。
形態・古生物の研究について。霊長類研究所では4年前からアジルテナガザル2頭、チンパンジー3頭の誕生が相次ぎ、彼らを対象とした身体プロポーション変化等の形態学的な発達研究に加え、血液中の成長ホルモン等の発達変化研究が継続している。さらにMRI、DEXA、pQCT等非侵襲的方法を用いた方法により、チンパンジーの咽頭構造の発達的変化をヒトと比較した発声機能の進化の研究、チンパンジーと比べヒトの乳児の突出した高体脂肪含量や四肢における骨塩量の形態発達学的変化等の研究がまとめられつつある。霊長類の化石発掘による人類進化研究は主としてアフリカでなされてきたが、東南アジアでの化石発掘研究も新たな地平を開きつつある。ミャンマーでの真猿類化石の発見、タイ国北部での大型類人猿化石の発見が相次いでいる。これらの発掘成果は真猿類や類人猿のアフリカ起源というこれまでの霊長類進化史説を書き換える可能性もあり将来にわたる継続的研究が期待される。
脳・認知科学研究では、上述のテナガザル2頭とチンパンジー3頭の未成熟個体について、統制のとれた実験条件下での発達研究が進展している。テナガザルでは身体的成長はマカクであるニホンザルとほとんど同じか少し早く、種々の行動の初出はニホンザルとチンパンジー中間に位置する。チンパンジーの発達研究では、24時間ビデオ撮影観察により、ヒトと同様な新生児微笑を示すこと、誕生直後はすべて母親依存であるが生後一年を過ぎる頃から子供達の間での遊びが始まること等、ヒトとの詳細な発達比較研究を今後も可能にする程の記録が集積できた。また、MRI等を用いた脳全体および脳各部位における容積の発達的増加等のデータも集積され、ヒトとの比較研究が期待される。
分子・遺伝の研究分野について。体毛、尿、糞、繊維質の食物を彼らが噛んで吐き出したシガミカス等から類人猿の遺伝子を収集する方法を検討してきた。これらの資料収集時に現地のトラッカー等の収集者や分析を担当する実験者からの汚染は避け難い問題であった。そこで対象とするDNA領域において、ヒトには存在し類人猿には存在しない制限酵素サイトを検索し、汚染されたヒトのDNAを、PCRを行う前に破壊しておき、汚染を除去する方法を開発した。また尿や糞を用いた類人猿の内分泌研究も発展した。これらは非侵襲的方法による生体資料収集として対象を霊長類以外にも広げることのできる非常に有用な方法である。また、霊長類研究所では15年来各種霊長類、動物からのDNAを精製し保存し続けており、その総数は3000個体を越え、霊長類DNA研究の拠点となっている。それらのDNA試料を用いた、性格関連神経伝達物質の遺伝子研究、進化の過程でゲノムに進入したレトロウィルス関連遺伝子の霊長類進化過程での変異研究、にも大きな進展があった。
これらの研究結果は平成15年3月13-15日に京都大学霊長類研究所共同利用ホミニゼーション(人類進化)研究会と共催で行ったシンポジウムで発表された。初日は犬山市の市民公開講演会とも共催し、研究代表者等による基調報告に続き、太陽系地球の進化、生命の多様性の獲得、人間性の由来等の講演が広く市民に開かれた形で開催した。後半二日間は本研究課題に関する報告会を霊長類研究所にて行った。
