蛋白質生合成系の拡張と非天然アミノ酸の導入による蛋白質の有機化学的機能拡張

研究概要	高効率4塩基コドンの使用による複数種類の非天然アミノ酸の同時導入 CGGUとGGGUの組み合わせにより高効率で複数種類の非天然アミノ酸が単一の蛋白質に組み込まれた。この方法で蛍光基と消光基をストレプトアビジンに組み込み、電子移動消光が起こることを、蛍光寿命測定によって確認した。新規蛍光性非天然アミノ酸の合成と蛋白質への導入ダンシル基などの蛍光基を側鎖にもつ非天然アミノ酸が新たに合成され、その内のいくつかは高効率で蛋白質に組み込まれることを確認した。またより大きな蛍光基でも長いスペーサーを介して結合すると蛋白質に導入可能であることを予備的な実験から確認した。これにより、非天然アミノ酸導入抗体や受容体による医療診断などの応用への道を拓いた。新規ペプチド核酸の開発天然のDNA以上にDNAやRNAと強くかつ配列特異的に対合する人工核酸を開発する目的で、新たに主鎖にピロリジン環を含むペプチド核酸が合成された。このペプチド核酸はピロリジン環の回りに2つの不斉炭素を持ち、計4種類の立体異性体が存在するが、アデニン側鎖をもつものについてはその4種類すべてを合成した。DNAとの相互作用を検討した結果、cis-L構造をもつものが最も高い親和性を示すことがわかった。ペプチド上での電子移動速度の測定非天然アミノ酸を導入して蛋白質中で電子移動を起こさせるための知見を得るため、ヘリックス構造をとるモデルペプチド上に電子供与基と受容基をもつ非天然アミノ酸を導入し、その間での光誘起電子移動を測定した。その結果、電子移動速度の推進力依存性はMarcus理論から予想される蛍光とよく一致することが明らかになった。またポリペプチド上での電子移動における電子移動再配向エネルギーは0.9eVであることが分かった。
