末端酸化酵素の基質酸化特異性とプロトン輸送能の研究

研究概要	大腸菌のbo型オキシダーゼや好熱性細菌caa3型オキシダーゼはSoxM型に属し、プラトン輸送活性を持つ。一方、SoxB型のもののアミノ酸配列の保存性はややひくいので、これらの型のオキシダーゼのについて次の研究をおこなった。 1。好熱性バシラス属細菌はcaa3型オキシダーゼとbo型のキノールオキシダーゼ以外にSoxB型に属するbo3型シトクロムcオキシダーゼを持つが、その存在量はすくない。B.stearothermophilus野生株K-1041に酵素のオペロンを持つプラスミドを形質転換したところ1-2nmol/mg proteinの本酵素が発現された。これをDEAEセルロースとハイドロキシアパタイトカラムにより純粋にした。 2。このbo3型オキシダーゼを大豆リン脂質と共にリポソーム小胞に再構成し、還元型シトクロムc-551を小量加えたところ、低いながらもH+ポンプ活性を検出することができた。 3。一方、caa3型シトクロムオキシダーゼbd型キノールオキシダーゼの両方を欠くmutantではこのbo3型オキシダーゼが主に働いているので、この休止菌を用いて嫌悪的条件でO2パルスをすることにより、H+/0を測定したところ5.0-5.3となった。これは野生株のそれ(6.0-6.5)よりやや低いが、4.0までは低下しないので、bo3型オキシダーゼは効率はひくいながらプロトンポンプであることが判明した。 4。bo3型オキシダーゼのESRスペクトルはCuAと高スピンヘムOと低スピンヘムBがSoxM型のオキシダーゼのものと類似している。またこのCuAに由来するg=2付近の微細装置はこの銅原子がN2O還元酵素と共通する構造を持つことを示した。RRスペクトルはヘムOが高スピン型、ヘムBが低スピン型であることを示すとともに、Feに配位するCOの配置がやや異なることを示した。 5。プラスミド上の遺伝子に部位特異性変異を導入してそれを発現して得た変異蛋白質を作成した。
