11q23転座型白血病の分子機構の解析

研究概要	MLL/MENはt(11;19)(q23;q13.1)の結果形成されるキメラ型転写因子遺伝子である。MLLはzinc finger typeの転写因子であり、MENはRNAポリメラーゼII転写伸長因子である。MEN及びMLL/MENのp53の機能に対する効果について検討を行った。まず、p53が転写を活性化する人工的レポーターRGC-Luc及びサイクリン Gプロモーターを用いたルシフェラーゼ・アッセイによりMLL/MENもMENもp53による転写活性化を抑制することを明らかにした。従って、MLL/MENはMEN部分依存性にp53の機能を抑制すると考えられた。さらに、免疫沈降・ウエスタン解析によりMLL/MENがMEN部分に依存してp53と結合することを証明した。欠失変異体も用いた実験により、MENはN末1/2の領域(p53結合領域)でp53と結合し、C末1/2の領域(p53抑制領域)でp53の転写活性化能を抑制していることが明らかになった。MENを遺伝子導入したRatl細胞のコロニー形成能が著名に増強することはすでに報告したが、p53結合領域あるいは抑制領域を欠いた欠失変異体においてはこのコロニー形成能の増強は観察されなかった。以上の結果より、MLL/MENはMEN部分でp53と結合し、その機能を抑えることにより、造腫瘍活性を発揮する可能性が示唆された。11q23転座による白血病発症の分子機構の多くは不明であるが、MLL/MENの場合には切断され短縮されたMLL部分ではなく転座相手遺伝子が白血病化に重要な役割を果たす可能性があると考えられた。
