MTH1遺伝子欠損マウスを用いた発がん研究

研究概要	電離放射線や環境中に存在する化学物質、さらには生体内での通常の代謝活動によっても活性酸素が生じている。これらは様々な作用を生体にもたらすが、中でもDNAの酸化は突然変異や発がんさらには生体の老化に深く関わっていることが示唆されてきた。種々のDNAの酸化的損傷の中で、グアニン塩基の酸化はその強力な突然変異原性により注目されている。活性酸素に伴うDNAの損傷と自然突然変異および発がんの関連を明らかにし、その過程を分子レベルで解明することを目的に、標的遺伝子組換えにより樹立したMTH1遺伝子の欠損マウスを用いて解析した。 (1)アニマルセンター内のSPF飼育条件下で1年6ヶ月後に剖検したところ、MTH1遺伝子欠損マウス(MTH1^<-1->)は野生型(MTH1^<+/+>)マウスに比べて肺などの特定臓器において、自然発生腫瘍頻度の上昇が観察された。このことは、MTH1蛋白質の欠損により、DNA中に取り込まれる変異原性ヌクレオチドである8-oxodGTPを排除する効率が低下することで、結果的に突然変異の上昇がもたらされ発がんの誘因になるという考え方を支持し、発がん抑制におけるHTH1蛋白質の重要性を示唆している。 (2)MTH1遺伝子欠損マウスの腺胃におて、H. pylori感染は、胃炎の持続と腺窩上皮の過形成を増強した。MTH1遺伝子欠損マウスでは特に、H. Pylori感染に伴って、異型度の高い病変(Atypical hyperplasia)の発生頻度が上昇した。 (3)近交系C57BL6/Jマウスに戻し交配が完了したMTH1遺伝子欠損マウスを用いて、New HITECマウスとの交配によって得られる仔マウスにおける突然変異を調べた。その結果、4週齢の脾臓から抽出したDNAの解析により、MTH1遺伝子欠損マウスの方が。野生型マウスに比べて、突然変異頻度が約2倍ほど上昇していた。 (4)MTH1遺伝子欠損マウスを用いて、X線によって誘発される腫瘍形成の実験を進めている。
