研究概要

バクテリオファージP1由来のCre-loxPシステムを利用し、lox71/66システムによるES細胞における遺伝子挿入法を確立した。また、このlox71/66のシステムとスペーサー部分の変異型lox511の両者の併用により、同じくES細胞の系で遺伝子の挿入をより効率よく行なうことに成功した。ES細胞を浮遊培養して胚様体へ分化を誘導する系をスクリーニングシステムとし、lox71を組み込んだ可変型トラップベクターを用いた遺伝子トラップ法により、24系統のトラップマウスを樹立した。19系統については、トラップした遺伝子の解析を行ない、これらが転写、細胞周期、翻訳、核と細胞質のタンパクの転送等に関与する遺伝子であることを明らかにした。作製したトラップマウスのうち、CBPおよびc-crkの破壊マウスについて詳細な解析を行なった。CBPでは、creb binding domainの3'側にトラップベクターが組み込まれており、トランケートされたタンパクが産生され、ドミナントネガティブ型の変異を生じることが示唆された。CBP破壊マウスのヘテロは、ヒト常染色体優性遺伝病であるRubinstein-Taybi症候群のモデルであり、ヒト患者で見られる症状や所見の殆どを有しており、よいモデルと考えられた。ホモマウスでは、血球及び血管形成の異常により、胚性9.5日目頃に致死となることが分かった。c-crk破壊マウスでは、F1およびF2では特に異常は認められなかった。しかし、C57BL/6へ戻し交配することにより、生後1日目に致死となることがわかった。人工細菌染色体のマウス受精卵への導入法を確立し、34系統とを樹立した。ま、Tミュータントおよびqkマウスに応用し、表現型をレスキューできることを実証した。
