大腸発がん過程におけるペロキシゾーム増殖活性受容体発現と化学予防

研究概要	大腸がん、大腸炎におけるPPARγの発現と関与の有無およびそのリガンドによる発がん抑制を以上の実験で試み、いくつかの成果を得た。 (1)雄性F344ラットに5%DSSを7日間飲水投与し、急性大腸炎を誘発した。同時にPPARγリガンド(troglitazone、30mg/kg体重)を連日計7回、強制胃内投与し、大腸粘膜における変化を組織学的に、PPARγ発現を免疫組織学的に検討した。その結果、PPARγはラット正常大腸粘膜上皮の表層部と増殖帯細胞の核に発現しており、DSS誘発急性大腸炎粘膜では正常大腸粘膜に比べ、PPARγの発現が増強していた。PPARγリガンド投与で大腸粘膜の炎症が軽減され、PPARγ発現が減弱した。 (2)Azoxymethane(AOM、20mg/kg体重、週1回計2回)とDSS(5%濃度で8週間飲水投与)でACFを誘発し、発がん剤誘発および炎症関連大腸発がんの初期過程におけるPPARγの発現を免疫組織学的に検討し、そのリガンド投与(troglitazone、0.05%混餌投与)によるACF発生の修飾作用を検討した。その結果、AFCでは背景粘膜に比べPPARγの発現が軽度増加し、PPARγリガンド投与で大腸がん前駆病変ACFの発がん有意(P<0.001、P<0.05)に抑制された(ACF数:AOM83±6、AOM+リガンド54±7、DSS35±13、DSS+リガンド11±12)。これらの材料におけるPPARγ(γ1、γ2)タンパク量やmRNAレベルの解析、PPARαやCOX-2発現の解析が進行中である。今回の結果ではPPARγリガンド投与でACF発生が抑制されたこと、特に炎症関連ACFの抑制は興味深い。発がん剤誘発・炎症関連大腸発がんモデルで、PPARγリガンドの長期投与やCOX-2阻害剤との併用投与で大腸がん抑制を試みる長期実験を準備している。
