ヒストンアセチル化とヒト白血病

研究概要	急性骨髄性白血病に見られるt(8;21)転座により生じるAML1-MTG8融合遺伝子をマウス骨髄系細胞株L-Gに導入すると、L-G細胞の分化は阻害され増殖が著しく促進され、その分化阻害は本来のAML1遺伝子の主要産物であるAML1bを発現させることにより可逆的に回復される。このことから、AML1bには好中球に分化させる機能があり、AML1-MTG8はAML1bと拮抗して作用することによって白血病発症の原因となると考えられた。AML1-MTG8によるL-G細胞の分化阻害に関与する機能ドメインを同定し、この領域には、新規に同定したMTG8のファミリータンパク質 MTGR1及びMTG16が結合することから、AML1-MTG8はMTGR1またはMTG16との複合体を形成して作用すると考えられた。さらに、AML1-MTG8はMTG16及びMTGR1との結合を介してヒストン脱アセチル化酵素と複合体を形成して作用し、転写を抑制することを示した。また、転写因子AML1はヒストンアセチル化酵素p300(CBP)と複合体を形成して転写を活性化することを明らかにした。一方、急性骨髄性白血病で見られたt(8;22)転座について解析し、アセチル化酵素をコードするMOZ遺伝子がp300遺伝子と融合して融合ヒストンアセチル化酵素が発現することを明らかにした。急性骨髄性白血病に見られるt(8;16)転座により生じるMOZ-CBP融合遺伝子産物はAML1やp53による転写活性化を著しく阻害することを示した。このように、白血病発症においてヒストンアセチル化の制御が重要であることが示された。
