c-kitレセプターチロシンキナーゼの恒常的活性化と腫瘍原性に関する研究

研究概要	c-kitレセプターチロシンキナーゼ(KIT)は、kinasc領域とjuxtamembranc領域の変異により、恒常的に活性化される。c-kit変異による二量体形成、恒常的活性化ならびに増殖シグナル伝達における細胞外領域、細胞膜貫通領域、細胞内チロシンキナーゼ領域の役割を検討するため、(a)juxtamembrane領域に変異を有するc-kit変異(KIT-G559)、kinase領域に変異を有するckit変異(KIT-V814)ならびに野生型c-kit(KIT-WT)の細胞外領域領域を欠失したDel-KITミュータント、(b)細胞外領域ならびに細胞膜貫通領域を欠失させ、細胞内領域とsrc N-terminal myristylation signalを結合させたsrc-KITキメラをそれぞれ作製し、IL-3依存性細胞株へ導入した。導入細胞において、細胞外を欠失させたDel-KIT-G559、Del-KIT一V814とも恒常的なチロシンリン酸化が認められIL-3非依存性増殖を促したが、Del-KIT-WTではほとんどリン酸化が認められず増殖刺激能も認められなかった。一方、src-KIT-G559、src-KIT-V814、src-KIT一WTでは、すべてのキメラともに恒常的リン酸化が認められたが、IL-3非依存性増殖はsrc-KIT-G559とsrc-KIT-V814のみに認められた。さらに、細胞外領域と細胞膜貫通領域を欠失したDel-KIT-G559、Del-KIT-V814、src-KIT-G559、src-KIT-V814は、細胞内領域で野生型KITと会合し共沈した。以上の結果より変異による恒常的活性化や下流へのシグナル伝達には、細胞外領域や細胞膜貫通領域は必須ではないと考えられた。
