癌細胞の増殖・浸潤に関与する活性酸素生成型NADPHオキシダーゼの活性化機構

研究概要	癌細胞の増殖・浸潤に関与する活性酸素生成型NADPHオキシダーゼの活性化には、低分子量G蛋白質Racとオキシダーゼ特異的シグナル伝達蛋白質(p47^<phox>,p67^<phox>とp40^<phox>)が移行して、細胞膜に存在する酵素本体シトクロムb_<558>(b_<558>)と相互作用する必要がある。平成11年度は以下のような成果を得た。 (1)Racの活性化を検出する新しい測定法を開発し、好中球刺激時のRacの上流で作用するシグナル分子を同定した。Racは、P67^<phox>のN末領域に存在するTPRドメインを介して作用し、オキシダーゼを活性化することを明らかにした。 (2)b_<558>と細胞質因子の相互作用は、p47^<phox>ドメインとp22^<phox>(b_<558>の小サブユニット)の細胞質領域との結合に依存するが、p47^<phox>C末領域の3つのセリン残基のリン酸化が、SH3ドメインのunmasking、p22^<phox>との結合、及びオキシダーゼ活性化能誘導に必要であることを示し、更にアラキドン酸がリン酸化と協同的に働いてp47^<phox>の構造変化を引起こしオキシダーゼを活性化することを明らかにした。 (3)p40^<phox>は休止時細胞でp67^<phox>と会合しているが、これは新規なドメイン間(それぞれPCモチーフとPB1ドメイン)の新しいタイプの蛋白質間相互作用によるものであることを示した。PB1ドメインは酵母のシグナル伝達蛋白質Bem1pにも存在し、その役割を明らかにするとと共に、その3次構造決定に成功した。 (4)p47^<phox>のN末領域に新規ドメイン(PB2ドメイン)を見い出し、その3次構造決定すると共に、PB2ドメインがPIP結合ドメインであり、p47^<phox>の膜移行に必須であることを明らかにした。
