研究概要	細胞外シグナルを直接核に伝達するJAK/STAT伝達系は、細胞増殖分化、がん化の決定に重要である。なかでもSTAT3はv-src、v-eykによる細胞がん化にも必須である。STAT3活性は、チロシンリン酸化による2量体形成、核移行に加えて、STAT3のS727リン酸化によっても転写活性能が制御されているが、今回IL-6刺激により活性化されるSTAT3キナーゼには、高濃度IL-6でのみ活性化されるMEK阻害剤感受性キナーゼ系と低濃度IL-6でも活性化される阻害剤H7に感受性のキナーゼ系があることを示し、後者のSTAT3キナーゼ系が、Ras/MAPK活性化の観察されない細胞や生理的濃度での刺激の際にも活性化されることから、実際に重要な役割を担うことを示唆した。さらにgp130受容体細胞内領域のうちSTAT3ドッキング領域として重要なpYXXQモチーフそのものが、H7感受性STAT3キナーゼ活性化に必要十分なモチーフであることを見い出した。種々の検討から、このpYXXQ由来伝達系を介したH7感受性STAT3キナーゼ伝達系は、Erk、p38、PKC8などのキナーゼ系とは異なるものであること、さらにH7感受性伝達路によるSTAT3のセリンリン酸化は核内でも認められることが判明した。今後、この新規伝達系の構成要素の同定、役割の解明が重要となる。また継続課題として、stat3遺伝子プロモーター上に存在する低親和性STAT3結合配列に変異を導入したプロモーターノックインマウスを作製し、STAT3によるstat3遺伝子活性化の役割などの解析を行いつつある。
