IL-15によるTh1細胞の選択的活性化機構とその癌免疫療法への応用

研究概要	IL-15によるTh1細胞の選択活性化機構を解析するために、H-2クラスIプロモーターと分泌型IL-15cDNAと非分泌型IL-15cDNAを用いてトランスジェニック(Tg)マウスを作製した。ともに胸腺でのT細胞分化は正常マウスとかわらなかったが、分泌型IL-15Tgマウスで脾臓、リンパ節でのメモリー型CD8T細胞が有意に増加していた。一方、非分泌型IL-15Tgマウスではトランスジーン由来のIL-15は細胞内に留まっているのみならず、内因性のIL-15mRNAの発現が抑制されていた。これらのIL-15TgマウスにSalmonellaを感染させて、CD4Th1細胞の産生をしらべたところ、分泌型IL-15Tgマウスで強いCD4Th1細胞の誘導が見られ、抵抗性を示した。分泌型IL-15Tgマウスではサルモネラ感染後、メモリー型CD8T細胞がIL15とIL-12によってby standerに活性化され、インターフェロンγを産生した。CD8T細胞の除去実験よりこのメモリー型CD8T細胞がCD4Th1細胞の強い誘導に重要であることがわかった。一方、非分泌型IL-15TgマウスではSalmonella感染後のIL-15の産生が認められず、またTh1細胞の産生が低く、菌の排除能も低下していた。IL-15はCD4Th1細胞の産生に重要なサイトカインと考えられる。IL-15の抗腫瘍効果を検討するために、IL-15遺伝子を導入したMethA線維肉腫をマウスの皮下または胸腔内に接種したところ、皮下接種ではおもにCD4T細胞が、胸腔内接種ではNK細胞とCD8T細胞がエフェクター細胞として活性化され、腫瘍を排除することが明らかとなった。
