熱・熱外中性子捕捉反応を利用した深在性放射線抵抗性腫瘍の治療に関する基礎研究

研究概要

1.熱外中性子照射時の硼素・中性子捕獲反応による棚素線量の水ファントム中のピーク深度はKUR、JRR4ビームで1.5-2.0cm、Petten炉で3cmであった。ピーク部と6cm深部での硼素線量比はPetten炉、KUR、JRR4で各々2.5、3.0、3.0以上であった。照射野(12cm)の中央をLiFプラスチック板で遮蔽すると熱中性子および硼素線量のピーク/6cm深部比は2.1-2.4に改善した。
2.BSHを投与したマウスの腫瘍に電撃処置を行うと腫瘍の硼素濃度は長時間高く維持され、BNCT効果が増強された。
3.BPAとBSHの両硼素化合物を同時併用するとBNCT時の細胞生存率曲線は単独の場合の中間に位置したが、腫瘍治癒を指標にすると相乗効果が認められた。
4.低酸素細胞の再酸素化はBPA-BNCTの場合はガンマ線照射の場合にほぼ類似し、BSH-BNCTでは中性子単独の場合の時間過程や程度に近い。
5.BPAの誘導体であるBPA-olのBNCT効果はBPA-ol>>硼酸=BPA>BSHの順であった。BPA-olの場合は、温熱処置(42.7℃-60分)によって更に大きくBNCT効果が増強された(約3倍)。
6.DMSOは熱中性子や熱外中性子の殺細胞効果から細胞を防護し、熱中性子で約1.9、熱外中性子で1.4の防護係数を示した。
