樹状細胞による抗腫瘍免疫応答の誘導

研究概要

樹状細胞は免疫応答の誘導において強力な抗原提示細胞として働くことが明かであり、欧米諸国をはじめとして日本国内でも抗腫瘍免疫応答の誘導において樹状細胞を"nature's adjuvant"として用いる可能性が検討されている。既に幾つかの報告ではその有効性が示されている。しかし、実際には当初期待されたほどでないのも事実である。その理由の一つとして、適用された樹状細胞が腫瘍特異的免疫応答を誘導するのに適したものでない可能性がある。また、実用に供するためには、汎用性が求められ、そのためには使用が限定される抗原ペプチドではなく、腫瘍由来の可溶化蛋白を用いる方向や、その樹状細胞への導入方法の検討がなされている。そこで、本研究では未熟樹状細胞の食作用能を用いたアポトーシス細胞捕食を介した腫瘍特異的免疫応答を誘導する可能性と樹状細胞を投与する場合の問題点を明らかにするための研究を行い、以下の結果を得た。
1.少数のアポトーシス細胞と混合培養した後に、捕食した樹状細胞の成熟度を調べてみたところ、アポトーシス細胞の捕食は検出されたが、その成熟度が極めて低いことが明らかになった。しかし、多数のアポトーシス細胞を加えた場合には、捕食樹状細胞自身がTNFαやIL-1を産生し、成熟することが示された。このようなサイトカイン産生は、培養中にアポトーシスからネクローシス状態になった細胞の捕食による可能性がある。しかし、アポトーシス細胞捕食後、成熟刺激を与えた場合には有効な抗原提示細胞となった。
2.未熟な樹状細胞を生体に投与した場合、所属リンパ器官への移動率ならびに生存率は成熟した樹状細胞を投与した場合に比べて低く、そのため、誘起される免疫応答誘も低いことが示された。これらの結果より、樹状細胞を有効に用いるためには、成熟した細胞を生体に投与することが重要と考えられる。
