IL-12の情報伝達とTh2サイトカインによる制御機構

研究概要

IL-12とIL-18の両者に応答するIL-12依存性T細胞株を用い、IL-12/IL-18によるIFN-g遺伝子発現に向けた情報伝達機構を解析し、次の結果を得た。
1.IL-12はSTAT4とMycを、IL-18はAP-1を誘導した。両者は単独では弱いレベルに、共刺激では高レベル(相乗的)にIFN-g遺伝子の転写を誘導した。
2.IL-12はMAPキナーゼのうちのERKのみを、IL-18はERKのみならずp38/JNKを強力に活性化し、AP-1がIL-18によって誘導されることが裏付けられた。
3.IFN-gmRNA発現及びIFN-g産生へのシグナル生成は、IL-12刺激よりもIL-18刺激が圧倒的に強力であった。又IFN-gプロモーター(約500bp)をルシフェラーゼ遺伝子に連結したレポーターを用いたルシフェラーゼアッセイにおいて、AP-1が最強の転写活性を示した。しかしながら重要なことに、AP-1の転写活性発現にはIL-12由来シグナルが必須であった。そのうちMycはAP-1転写活性化を有意に増強した。又、マウスIFN-gプロモーターではAP-1上流にSTAT4結合部位が存在しないことに加えて、AP-1下流のプロモーターに対してもIL-12とIL-18の相乗効果がみられたことより、STAT-1はAP4下流に作用することが示唆された。
以上、IL-12シグナルは、強力な転写因子AP-1の機能発現に必須の転写因子を作用させることによってIFN-g遺伝子発現に貢献することが示唆された。
