多剤耐性の分子標的としてのABCトランスポーター遺伝子群の発見と機能

研究概要	生体内の様々な器官や組織においてABCトランスポーターは発現しており異物・毒物排出ポンプとしての生理的に重要な役割を担っている。がんにおける抗がん剤の感受性を制御する分子標的としての臨床診断へ利用するためにはABCトランスポーターのがんにおける特異的発現の分子的機構を把握することが必須である。我々は 1.P-糖蛋白質の遺伝子MDR1遺伝子がヒトがんで発現亢進する特異的な分子機序について(1)MDR1プロモーターのCpGサイトのメチル化の有無(2)ゲノム不安定性に伴う遺伝子再編成機構(3)転写因子YB-1の核内局在による発現上昇を明らかにした。また、膀胱癌や急性骨髄性白血病、肺癌などの臨床腫瘍でいずれかの機構によりMDR1遺伝子の発現亢進を明らかにしてきた。 2.MRPファミリーのMRP2/cMOATならびにMRP3については、(1)遺伝子導入実験によりビンクリスチンやシスプラチン、エトポシドの耐性に関与すること。(2)大腸癌ではMDR1,MRP3の発現は癌部にて発現の低下が認められたが、MRP2の発現は逆に亢進していることを明らかにした。(3)肝炎ウイルス感染患者ではMRP2の発現が有意に低下していることを示した。以上より、臓器特異的な抗がん剤耐性獲得機構を正しく把握して、がんに伴う遺伝子変化やエピジェネティックな変化を標的とした治療法に繋げたいと考えている。
