抗癌剤で活性化される転写因子を標的とした薬剤開発に向けた基礎研究

研究概要

研究計画は、1.プロモーター解析と2.転写因子解析の2本柱でスタートしたが、前者については新規プロモーター単離と機能解析で大きく進展できたが、後者については既知の転写因子解析のみにとどまり、若干の不満があるものの新しい発見がいくつかあり今後の課題として残された。平成11年度明らかにした点は以下である。1.(1)損傷DNAを認識するHMG1の遺伝子プロモーター構造及び、シスプラチン耐性細胞での過剰発現の分子を明らかにした。(2)シスプラチン耐性細胞で過剰発現しているSTAT3及びIL13-Rα1遺伝子のプロモーター構造及び、転写制御について解析した。(3)薬剤耐性遺伝子としてMRP2のプロモーターの構造機能解析を行った。2.(1)YB-1の発現を臨床試料(卵巣癌42例、大腸癌26例)で解析し、増殖や予後との関連を明らかにした。(2)抗癌剤でNF-kBが活性化されること、AP-1がグルタチオン関連のレドックス制御を受けること、シスプラチン耐性細胞でCTF/NF-1が過剰発現していることを明らかにした。(3)YB-1やHMG1が癌抑制遺伝子p53と分子会合することを発見し、その意義について現在検討している。今後はプロモーター解析による新規転写因子のクローン化による同定と既知の転写因子を含めた蛋白・蛋白会合の核内マップを作成することが、増殖、アポトーシス、耐性化との関連から最も有力な分子標的を明らかにしていく上で大切と考えられる。現在、酵母ワンハイブリッドシステムの導入を進めており、新規の転写因子の同定に向けて研究を継続している。更に、抗癌剤で発現誘導される遺伝子の単離を目指して、ディファレンシャルディスプレイ法を行い新しい遺伝子の同定も進めており、そのプロモーター解析も着実に進展している。
