分子シャペロンによる遺伝子病治療の試み

研究概要	がんや脳疾患などの後天的な病気、および先天的な遺伝病の多くは、重要な機能を持つ蛋白質の点突然変異や部分的欠失で起こる遺伝子病といえる。これらの変異によってその蛋白質は正しく折れたたまれなくなり、機能が低下したり異常な凝集体を形成するようになることで病気が発症する。分子シャペロンは蛋白質の正しい折りたたみに必須の機能を持つ。最近、変異蛋白質であっても分子シャペロンが過剰に存在すると正常に機能するようになることがわかってきた。分子シャペロンによる遺伝子病治療の基礎研究として、今年度は以下の点について検討した。 1.球脊髄性筋萎縮症(SBMA)はCAG triplet repeat病の1つであり、アンドロゲン受容体(AR)のCAG(Glutamine) repeatが異常に伸長していることが原因である。この変異ARは細胞内では凝集体を形成するが、分子シャベロン(Hsp70,Hsp40)によってこの凝集体形成が抑制されることを示した。 2.Epstein-Barrウィルスの蛋白質(BZLF1)を昆虫細胞で過剰発現させると、不溶性の凝集体を形成するが、このとき分子シャペロン(Hsp70,Hsp40)を同時に発現すると、可溶化される蛋白質が数倍程度増加する。 3.分子シャペロンのHSP40(DNAJ)相同体のうち、まだ未知の11個の新規cDNAをクローニングした。
